Some Properties of Nicotinamide Adenine Dinucleotide Glycohydrolase from Human Nasal Polyp

Abstract
Some properties of nicotinamide adenine dinucleotide (NAD) glycohydrolase in cell-free extracts of nasal polyp have been studied.
The enzyme shows a sharp optimum pH at 6.8, and exhibits a characteristic heat lability, being activated by heat-treatment at 45¡Æ for 10 minutes and inactivated rapidly over 45¡Æ.
The enzyme is markedly inhibited by nicotinamide, the concentrations of which causing: 50% and 100% inhibition are 2 and 10 mM, respectively. In contrast to nicotinamide, nicotinic acid is without effect even at 25 mM. On the other hand, 3-egeijlbyiidlibi enhances rather the hydrolytic activity of the enzyme.
The enzyme is found to be "isonicotinic acid hydrazide-insensitive", being not inhibited by, isonicotinic acid hydrazide but forming isonicotinic acid hydrazide of NAD.
The activation by heat at 45¡Æ of the enzyme suggests that some heat-labile inhibitor of protein nature is associated in vivo with the enzyme.TG
